What it is
Retatrutide is an investigational peptide developed by Eli Lilly. It is a triple agonist: it simultaneously activates three receptors involved in glucose and energy metabolism — GLP-1, GIP (glucose-dependent insulinotropic polypeptide), and the glucagon receptor.
How it differs from semaglutide
Where semaglutide activates only GLP-1, retatrutide’s additional GIP activation may improve insulin sensitivity, and glucagon activation may increase energy expenditure (resting metabolic rate). In phase 2 trials, retatrutide produced larger average body weight reductions than any GLP-1-only agent published to date.
Regulatory status
As of 2026, retatrutide is in late-stage clinical trials and has not been approved by any major regulator. It is not yet available as a pharmaceutical product.
